A special cutting tool is run over the dough just before baking to prick any bubbles which might make the matza puff up; this creates the familiar dotted holes in the matzo. After the matzos come out of the oven, the entire work area is scrubbed down and swept to make sure that no pieces of old, potentially leavened dough remain, as any stray pieces are now chametz, and can contaminate the next batch of matzo. Some machine-made matzos are completed within 5 minutes of being kneaded.

The tertiary amines on the interior of the dendrimer can also participate in the buffering activity, causing the molecule to puff up; additionally, as the PAMAMs take on more and more positive charge, fewer of them are required for the optimal PAMAM-DNA interaction, and free dendrimers are released from the complex. Dendrimer release and swelling can eventually lyse the endosome, resulting in release of the cargo DNA. The activated PAMAM dendrimers have less spatial barrier to interior amine protonation, which is thought to be a major source of their advantage over non-activated PAMAM.

The Fool tells this to his friends, and while they spot the penguins, they are in a dilemma as they do not know how to get the penguins from the South Pole. The Runner suggests that he should complete this task by running to the South Pole and then bring the penguins back to Russia, but the sharp shooter objects to this, because he had already used his last bullet to wake the runner up, so if the runner were to fall asleep like last time, he couldn't be awoken. Suddenly, the Puffer wakes up from his slumber and tells the others that he is going to puff up and therefore, he transforms into a strong heavy set man. The Puffer realizes that if they couldn't go to the South Pole, then he'd have to bring the South Pole to them.
