Dark field illumination uses a carefully aligned light source to minimize the quantity of directly transmitted (unscattered) light entering the image plane, collecting only the light scattered by the sample. Dark field can dramatically improve image contrast – especially of transparent objects – while requiring little equipment setup or sample preparation. However, the technique suffers from low light intensity in final image of many biological samples and continues to be affected by low apparent resolution.

Individual images of stained or unstained particles are very noisy, and so hard to interpret. Combining several digitized images of similar particles together gives an image with stronger and more easily interpretable features. An extension of this technique uses single particle methods to build up a three-dimensional reconstruction of the particle. Using cryo-electron microscopy it has become possible to generate reconstructions with sub- nanometer resolution and near-atomic resolution first in the case of highly symmetric viruses, and now in smaller, asymmetric proteins as well.

This so-called silver chromate impregnation procedure stains entirely or partially the cell bodies and neurites of some neurons -dendrites, axon- in brown and black, allowing researchers to trace their paths up to their thinnest terminal branches in a slice of nervous tissue, thanks to the transparency consequent to the lack of staining in the majority of surrounding cells. Modernly, Golgi- impregnated material has been adapted for electron-microscopic visualization of the unstained elements surrounding the stained processes and cell bodies, thus adding further resolutive power.

Digital holographic microscopy thus makes it possible to visualize and quantify transparent objects and is therefore also referred to as quantitative phase-contrast microscopy. Traditional phase contrast or bright field images of living unstained biological cells, Figure 3 (right), have proved themselves to be very difficult to analyze with image analysis software. On the contrary, phase shift images, Figure 3 (left), are readily segmented and analyzed by image analysis software based on mathematical morphology, such as CellProfiler. 3-dimensional information An object image is calculated at a given focal distance.

Traditionally, Whitland is seen as the site of an assembly of lawyers and churchmen, sometimes described as the first Welsh parliament, called in 930 by King Hywel Dda to codify the native Welsh laws. Whitland takes its name from its medieval Cistercian abbey. The monastery pre-dates Tintern but now is very much a ruin. In Literary Theory: An Introduction (1983, revised 1996), Eagleton surveys the history of theoretical approaches to literature, from its beginnings with Matthew Arnold, through formalism, psychoanalysis, and structuralism, to post-structuralism. In the process, he demonstrates what is the thesis of the book: that theory is necessarily political. Theory is always presented as if it is unstained by point of view and is neutral, but in fact it is impossible to avoid having a political perspective. The phase contrast between the two rays can be detected with the human eye using particular optical filters.

He is known for line of Gennari, a macroscopically white band seen in the cerebral cortex of the occipital lobe, which he observed on 2 February 1776 in the course of examining frozen sections of unstained human brain during his study in medical school. He mentioned it in his book De peculiari structura cerebri, nonnulisque ejus morbis, and referred to it as lineola albidior. The discovery of this part of the brain is considered the first evidence that the cerebral cortex was not uniform in structure.

Proponents claim that live blood analysis provides information "about the state of the immune system, possible vitamin deficiencies, amount of toxicity, pH and mineral imbalance, areas of concern and weaknesses, fungus and yeast." Some even claim it can "spot cancer and other degenerative immune system diseases up to two years before they would otherwise be detectable" or say they can diagnose "lack of oxygen in the blood, low trace minerals, lack of exercise, too much alcohol or yeast, weak kidneys, bladder or spleen." Practitioners include alternative medicine providers such as nutritionists, herbologists, naturopaths, and chiropractors. Dark field microscopy is useful to enhance contrast in unstained samples, but live blood analysis is not proven to be useful for any of its claimed indications.

When observing an unstained biological specimen, the scattered light is weak and typically phase-shifted by −90° (due to both the typical thickness of specimens and the refractive index difference between biological tissue and the surrounding medium) relative to the background light. This leads to the foreground (blue vector) and background (red vector) having nearly the same intensity, resulting in low image contrast. In a phase-contrast microscope, image contrast is increased in two ways: by generating constructive interference between scattered and background light rays in regions of the field of view that contain the specimen, and by reducing the amount of background light that reaches the image plane. First, the background light is phase-shifted by −90° by passing it through a phase-shift ring, which eliminates the phase difference between the background and the scattered light rays.

For the next 80 or 90 years succeeding the banishment of the apostle John to Patmos covering the successive reigns of the emperors Nerva, Trajan, Hadrian and the two Antonines (Antoninus Pius and Marcus Aurelius), a golden age of prosperity, union, civil liberty and good government unstained with civil blood unfolded.
