The umu test, using only a single Salmonella strain, could potentially test a greater range of new chemicals with the same resources. Nevertheless, additional strains have been developed in order to increase sensitivity for the detection of genotoxins, belonging to certain classes, such as nitroarenes. All umu strains carry the plasmid pSK1002 which bears an umuD gene and an umuC gene fused with lacZ, the structural gene for beta- galactosidase. The induction of mutator gene umuC by DNA-damaging agents is detected by measuring intracellular beta-galactosidase levels. Like other tests, the strains have been genetically modified further.

The colonial-era sources are not entirely clear or in agreement about the nature of the structure of the Inca government. However, its basic structure can be spoken of broadly, even if the exact duties and functions of government positions cannot be told. At the top of the chain of administration sat the Sapa Inca. Next to the Sapa Inca in terms of power may have been the Willaq Umu, literally the "priest who recounts", who was the High Priest of the Sun.

The specific type of damage is determined by the size of the colonies, distinguishing between genetic mutations (mutagens) and chromosomal aberrations (clastogens). The SOS/umu assay test evaluates the ability of a substance to induce DNA damage; it is based on the alterations in the induction of the SOS response due to DNA damage. The benefits of this technique are that it is a fast and simple method and convenient for numerous substances. These techniques are performed on water and wastewater in the environment.

Filipinos brought vinegar, bagoong, and lumpia, and during the 20th century immigrants from American Samoa brought the open pit fire umu and the Vietnamese introduced lemongrass and fish sauce.
Throughout the video, misono is shown to be a humanized version of a dog who adores their owner and waits for them to come home when they leave. The song was more rock-oriented than her previous singles, showing a genre direction misono would continue to use for the rest of her career.
It is cooked very slowly in an umu for 12 to 24 hours. The plant is usually associated with the far north of New Zealand but may have been grown further south during the warmer climatic period associated with early Polynesian settlement. A geophysics study showed that it was only one of six such pits in a rough horseshoe shape on the edge of a lagoon. The study also showed the site to be much bigger than previously thought—at least and possibly larger as two boundaries have not been accurately plotted.
Considered the staple starch of traditional Polynesian cuisine, taro is both a common and prestigious food item that was first introduced to the Polynesian islands by prehistoric seafarers of Southeast Asian derivation. Inti, as represented by José Bernardo de Tagle of Peru The Sapa Inca was conceptualized as divine and was effectively head of the state religion. The Willaq Umu (or Chief Priest) was second to the emperor. Local religious traditions continued and in some cases such as the Oracle at Pachacamac on the Peruvian coast, were officially venerated. Following Pachacuti, the Sapa Inca claimed descent from Inti, who placed a high value on imperial blood; by the end of the empire, it was common to incestuously wed brother and sister.
The following day, the Spaniards resisted several Inca counterattacks and mounted a renewed assault at night using scaling ladders. In this way, they captured the terrace walls of Sacsayhuamán while the Inca army held on to the two tall towers of the complex. The Inca commanders, Paucar Huaman and the high priest or Willaq Umu, decided to leave the confinement of the towers and fight their way towards Calca, the site of Manco Inca's headquarters, to bring back reinforcements. The attempt was successful and the towers were left under the command of Titu Cusi Gualpa, an Inca nobleman.

There have been numerous accounts on the life and person of Iguedo. Her history seems to have been a greater puzzle to Historians than that of Eri. However, none of the accounts about her can be out-rightly and correctly accepted or rejected as some of the data came purely through oral tradition and scanty archaeological discoveries although there is near unanimity in the various accounts concerning the strong connection between an individual called Iguedo and the towns that constitute Umu-Iguedo clan. One of the more accepted opinions hold that she was a daughter of Eri.
Cordyline pumilio was cultivated by Māori in the Waikato district and elsewhere all over New Zealand. Young seedlings were carefully selected and planted out, and after perhaps three years the roots were dug up, stacked in small piles, and dried in the sun. As they dried, the fibrous roots were burned off, and then the roots were scraped and baked slowly in an umu or hāngi, requiring twelve to eighteen hours to cook. The cooked roots were chewed, or pounded and washed and squeezed to extract the sugar, which was eaten with fern root as a relish.
A Samoan 'umu at the early stage of heating the rocks An earth oven, ground oven or cooking pit is one of the simplest and most ancient cooking structures. At its most basic, an earth oven is a pit in the ground used to trap heat and bake, smoke, or steam food. Earth ovens have been used in many places and cultures in the past, and the presence of such cooking pits is a key sign of human settlement often sought by archaeologists. Earth ovens remain a common tool for cooking large quantities of food where no equipment is available.

Finally, Nnamovo, a man who was believed to have come from Onitsha married Iguedo and she gave birth to Nando. It was in the land founded by Nando that Iguedo died and was buried. However, these attempts to credit Iguedo as the maternal ancestry of Umuleri town seems to contradict the widely accepted knowledge that Iguedo was a daughter of Eri as there was no way She could be a daughter of Eri and still married him or his son in order to beget Umuleri.
The stems and fleshy rhizomes of C. australis are high in natural sugars and were steam-cooked in earth ovens (umu tī, a large type of hāngi) to produce kāuru, a carbohydrate- rich food used to sweeten other foods. The growing tips or leaf hearts were stripped of leaves and eaten raw or cooked as a vegetable, when they were called kōuka—the origin of the Māori name of the tree. The southern limit of kumara (sweet potato) cultivation was at Banks Peninsula at 43°S, and south of there a culture developed around C. australis. Natural and planted groves of the cabbage tree were harvested.
Kāuru could be stored dry until the time came to add it to fern root and other foods to improve their palatability. The sugar in the stems or rhizomes would be partially crystallised, and could be found mixed in a sugary pulp with other matter between the fibres of the root, which were easily separated by tearing them apart. Kāuru could also be dipped in water and chewed, and was said to smell and taste like molasses. Evidence of large cooking pits (umu tī) can still be found in the hills of South Canterbury and North Otago, where large groves of cabbage trees still stand.
An elaborate system of rocky platforms were found in the caves, constructed mainly with pieces of fallen lava-rock and built up to a height of about above the cave floor. Freeman recorded 152 platforms, 129 in the north cave, and 23 in the south cave. He also found numerous umu cook sites, fireplaces, and kitchen-middens as well as several lumps of ele a type of red volcanic rock used as a natural dye for Samoan siapo or tapa, a traditional bark cloth material. He found 5 stone adzes, four in the North Cave and one in the South Cave, a common type of prehistoric adze found in Samoa.

Popular as a "grass hut" or beach fale in village tourism, many are raised about a meter off the ground on stilts, sometimes with an iron roof. In a village, families build a faleo'o beside the main house or by the sea for resting during the heat of the day or as an extra sleeping space at night if there are guests. The tunoa (cook house) is a flimsy structure, small in size, and not really to be considered as a house. In modern times, the cook house, called the umukuka, is at the rear of the family compound, where all the cooking is carried out in an earth oven, umu, and pots over the fire.
As soon as the death has occurred all family members will be notified, nowadays often by a radio message and they are supposed to come to the putu (funeral rites), no excuses. For friends or distantly related members it is enough that they come, pay their respect to the dead, bring a small gift for the widow (or whatever the case may be), have their share of food and then leave until the actual burial. The household of the deceased is supposed to provide a meal, or meals if it takes long, to all mourners. In case of a large family this is a huge and expensive operation with big umu, and tons of food.

It is understood by the carbon dating of Umu (ovens) that the Maori people settled in this region some 700 years ago. The district takes its name from the Buller River, itself named for Charles Buller, Member of Parliament in the United Kingdom of Great Britain and Ireland (UK) and director of the New Zealand Company, a UK-based company in the early 1800s with a royal charter supporting colonization efforts of New Zealand. During the period 1853 to 1876, the current area of Buller District was administered as part of Nelson Province. Generally, the sasa is performed by a large group of people, it is normally performed sitting down, but there are parts of the dance which require the group to stand up. The movements depict everyday life, from the movement of fish in the water, to the flying birds in the sky, from cooking the umu to cleaning the house, and even a form of voyaging, where the group move into the form of a large canoe, having the arms on the outside mimic the movement of paddles in the water. Every Sasa is different, some movements have never changed, however nowadays, more contemporary moves are now being added to the Sasa. The Samoa 'ava ceremony is always included in the Sasa where the group would mimic the Taupou making 'ava.
The umu operon is useful as it is likely to be more involved in mutagenesis than the other known SOS genes (Kato and Shinoura, 1977), and can be taken advantage of in the same way as the SOS Chromotest. Taking advantage of the operon fusion placing the lac operon (responsible for producing beta-galactosidase, a protein which degrades lactose) under the control of an SOS-related protein, a simple colorimetric assay for genotoxicity is possible. If a genotoxin is present, the resulting DNA damage will result in the production of beta- galactosidase instead of DNA repair as a result of the gene fusion. A lactose analog is added to the media which is degraded by the beta-galactosidase, producing a colored compound which can be measured quantitatively through spectrophotometry.
Eyre foresaw ongoing problems with Māori and to remedy the situation, he instructed land purchase commissioner Walter Mantell and surveyor Alfred Wills to define the native reserves after the winter. Māori chiefs north of the Waimakariri River disputed that their land had ever been sold and demanded a corridor wide along the Waimakariri all the way to the Southern Alps, plus all the coastal land between the Waimakariri and the Ashley. Mantell granted them a reserve of at Tuahiwi, the pa site at Kaiapoi, and at a pa on the north bank of the Waimakariri. South of that river, reserves set aside were at Taumutu near the outlet of Lake Ellesmere / Te Waihora (), at Arowhenua (south of Temuka; ), on the Temuka River (Te Umu Kaha River; ), at Caroline Bay (later part of Timaru; ), and on the north bank of the Waitaki River ().

Moreover, it matches the cycle of rotation of the stars around the pole star, the real reason being that the Earth rotates at a constant angular velocity. If hours were to represent divisions of the uniform rotation, they must also be uniform, and not be variable. There were two days of the year when all 24 hours were of the same length: the two equinoxes. The standard double hour (beru), of equinoctial length, representing two modern hours, of which there were 12 in the standard day (umu), was not conceived as being one of day and one of night, but as being just two consecutive equal-length hours. One standard day thus went on to become two consecutive equal 12-hour clockfaces in modern clock time. 30 standard days were a standard month, and 12 of those a standard year of 360 days. Some juggling of month lengths to make the 12 months fit the year was still required. Within a day, single hours were unreliable.
The mascapaicha was the mark of absolute imperial power, to the point that "to take" or "to gird the mascapaicha" or "to claim the scarlet fringe" was the periphrase in use to signify the coming of the new emperor or Sapa Inca, during a ceremony where he received it from the hands of the Willaq Umu, the high priest or pontiff of the Inca religion. One of the stories told about Sinchi Roca was that during his reign a llama shepherd had entered his house and took a young woman whom he cherished very well. The Inca then had them captured and tortured him to tell why he had committed such a sin; the young woman, not wanting to see the villager tortured anymore, revealed that she had fallen in love with the llamamichi (shepherd of llamas) as soon as he saw him wearing a wacanqui (amulet of love), and the shepherd confessed, in turn, that he had received such an object from a demon in a cave.
