The duration can last from 4 to 12 hours depending on route of administration, dose, and other factors. With insufflation, the effects are more abrupt and intense but have a significantly shorter duration, while oral usage results in a milder, longer experience. When insufflated, the onset happens very rapidly, usually reaching the peak at about 20–40 minutes and plateauing for 2–3 hours. 2C-B is also considered one of the most painful drugs to insufflate, with users reporting intense nasal burning.

Dextroamphetamine is also used recreationally as a euphoriant and aphrodisiac, and like other amphetamines is used as a club drug for its energetic and euphoric high. Dextroamphetamine is considered to have a high potential for misuse in a recreational manner since individuals typically report feeling euphoric, more alert, and more energetic after taking the drug. Large recreational doses of dextroamphetamine may produce symptoms of dextroamphetamine overdose. Recreational users sometimes open dexedrine capsules and crush the contents in order to insufflate it or subsequently dissolve it in water and inject it.

Any damage to the inside of the nose is because cocaine highly constricts blood vessels – and therefore blood and oxygen/nutrient flow – to that area. Nosebleeds after cocaine insufflation are due to irritation and damage of mucus membranes by foreign particles and adulterants and not the cocaine itself; as a vasoconstrictor, cocaine acts to reduce bleeding. Rolled up banknotes, hollowed-out pens, cut straws, pointed ends of keys, specialized spoons, long fingernails, and (clean) tampon applicators are often used to insufflate cocaine. Such devices are often called "tooters" by users.

The beans have been found to contain up to only 0.04% 5-MeO-DMT and 0.16% DMT. The leaves and bark also contain small amounts of DMT, 5-MeO- DMT and related compounds.Schultes 1976 Schultes 1977 Pachter et al. 1959 At up to 0.04% (0.4 mg per gram) 5-MeO-DMT, an effective light 5 mg dose of insufflated 5-MeO-DMT would require over 12 grams of beans. It would be extremely difficult to insufflate such a quantity, as tolerance would likely develop before the 12-gram nasal intake could be completed.

Individual sensitivity to 5-MeO-DMT varies. It has been documented that the threshold dose in some individuals is as much as 10 mg insufflatedShamanic Snuffs or Entheogenic Errhines by Jonathan Ott, Page 102, 2001, requiring over 24 grams of beans for an effective dose of 5-MeO-DMT. At up to 0.16% (1.6 mg per gram) DMT, an effective 40 mg dose of insufflated DMT would require 25 grams or more. Because of its volume, it’s likely to be impossible to insufflate the 25 grams of beans required to reach the active dose of DMT present in the beans.

The use of diluents in illegal drugs reduces the potency of the drugs and makes it hard for users to determine the appropriate dosage level. Diluents include "foodstuffs (flour and baby milk formula), sugars (glucose, lactose, maltose, and mannitol), and inorganic materials such as powder." The diluents used often depend on the way drug purchasers consume particular drugs. Drug dealers selling heroin to users who inject dilute the drug with different products from dealers selling to users who smoke or insufflate the drug; diluents which can easily form a solution with water for injecting heroin can be problematic for users who are sniffing the powder.
