Two iconic figures are introduced in Season 2: Frank Castle aka The Punisher (Jon Bernthal), who functions as a moral mirror for Matt, and Elektra Natchios (Elodie Yung), a deadly woman from Matt's past who was also trained by Stick and arrives in Hell's Kitchen seeking Matt's help.

The film's main character, Miyuki Minazuki, ranked number three in The Assassins Guild, known as the "Stray Cat". A stoic and deadly woman, she is forced to deal with the crumbling Guild, and crazy state of affairs, seeking out the cause of all this, "Hundred Eyes". She lives in a Japanese home with her grandmother, and her weapon is a pistol. Former number one, now crippled and retired to an honorary ranking of number zero "The Champ" Goro Hanada is an acquaintance of Stray Cat's, offering news and unsolicited advice.

Isha katlanit (, literally: "lethal/deadly woman") is used in halakha ("Jewish law") for a married woman who has become a widow twice. Such a woman, it is said, should not marry again, because marrying her carries the risk that her next husband may also die (i.e., she will become the "cause" of his death because her marriage to her two previous husbands ended when they died.) The origin of this rule is Talmudic. There is a dispute in the Talmud about whether a woman becomes a katlanit ("causing death") after the death of two husbands or the death of three husbands.

After the "Cocaine Coup" in 1980, installed by various Bolivian drug kingpins, Minister of the Interior Luis Arce Gomez put Atala in charge of smuggling cocaine and money laundering operations across the country. She paid her smugglers $20,000 monthly with a $500 tax for each kilogram of cocaine. According to ex-DEA agent Michael Levine (later her undercover partner), Atala was described as a "very beautiful but deadly woman" who could order people dead anywhere in the country. Her mansion in Santa Cruz was known as the "Torture House" due to her enforcers taking in enemies and torturing them.

Prior to her infestation, Kerrigan is described as being a graceful and deadly woman, exceedingly agile and athletic, possessing jade- green eyes and brilliant red hair usually worn as a ponytail. The novel Queen of Blades describes her facial features as being too strong to be classified as beautiful, but instead as striking and completely fitting for her personality. Kerrigan is rarely seen out of her armor, a form-fitting hostile environment suit specifically designed for Ghost operatives and equipped with a personal cloaking device, but when off-duty, she is described as wearing a soft work shirt, worn cotton pants with a dusty leather jacket, and high leather boots. Even then, it is uncommon for Kerrigan to be unarmed: she is always equipped with at least a combat knife.
