The most commonly reported adverse effect of taking a misoprostol by mouth for the prevention of stomach ulcers is diarrhea. In clinical trials, an average 13% of people reported diarrhea, which was dose-related and usually developed early in the course of therapy (after 13 days) and was usually self-limiting (often resolving within 8 days), but sometimes (in 2% of people) required discontinuation of misoprostol. The lead compound from Trius, tedizolid phosphate (formerly torezolid phosphate, TR-701), successfully completed its first Phase 3 clinical trial (ESTABLISH 1) in acute skin and skin structure infections (ABSSSI). It is a once daily, IV and oral, oxazolidinone being developed for the treatment of serious grampositive infections, including those caused by methicillin-resistant Staphylococcus aureus (MRSA). The Phase 3 trials are being conducted under a Special Protocol Agreement with the US Food and Drug Administration and are using the draft FDA guidance of cessations of lesion spread at 48–72 hours as the primary endpoint. In the ESTABLISH 1 Study, 200 mg of Tedizolid given for a short course of therapy (six days) was shown to be safe, well tolerated and was as effective as the traditional length of therapy with linezolid (600 mg for ten days).

Jacobson and Truax later defined clinical significance as “the extent to which therapy moves someone outside the range of the dysfunctional population or within the range of the functional population.” They proposed two components of this index of change: the status of a patient or client after therapy has been completed, and “how much change has occurred during the course of therapy.” Clinical significance is also a consideration when interpreting the results of the psychological assessment of an individual. Frequently, there will be a difference of scores or subscores that is statistically significant, unlikely to have occurred purely by chance.

Grünbaum argues that Freud, in a 1917 lecture on "Analytic Therapy", advanced a defense of psychoanalysis as a method of clinical investigation that went unnoticed in scholarly literature until Grünbaum drew attention to it in papers published in 1979 and 1980. Grünbaum refers to this defense as the "Tally Argument", and maintains that Freud used it to justify the claim that durable therapeutic success guarantees that the interpretations made in the course of therapy are accurate.
